چکیده

wnloaded mismatch repair (MMR) pathway is involved in the removal of DNA base mismatches that arise during DNA replication or are caused by DNA damage. Mutations in four genes involved in MMR, , MLH1, PMS2 and MSH6, predispose to a range of tumorigenic conditions, including hereditary lyposis colon cancer, also known as Lynch syndrome. Here we discuss the canonical MMR pathway e burgeoning evidence for noncanonical roles for the MMR genes, and highlight the therapeutic ations of MMR. In particular, we discuss how the DNA repair defect in MMR-deficient cancers could implic be exploited by the development of novel therapeutic strategies based on synthetic lethal approaches.

Involvement of ERCC1/XPF and XPG in oligodeoxynucleotide-directed gene modification.

Oligodeoxynucleotide (ODN)-mediated gene alteration was postulated to occur in two steps, DNA strand pairing and DNA repair. Once alignment has occurred through homologous strand pairing, a single mismatch is formed between an oligonucleotide and one of the target strands. Because of this mismatch, it has been suggested that proteins involved in a mismatch repair pathway (MMR) participate in th...

DNA Mismatch Repair and Oxidative DNA Damage: Implications for Cancer Biology and Treatment

Many components of the cell, including lipids, proteins and both nuclear and mitochondrial DNA, are vulnerable to deleterious modifications caused by reactive oxygen species. If not repaired, oxidative DNA damage can lead to disease-causing mutations, such as in cancer. Base excision repair and nucleotide excision repair are the two DNA repair pathways believed to orchestrate the removal of oxi...

Active DNA demethylation mediated by DNA glycosylases.

Active DNA demethylation is involved in many vital developmental and physiological processes of plants and animals. Recent genetic and biochemical studies in Arabidopsis have demonstrated that a subfamily of DNA glycosylases function to promote DNA demethylation through a base excision-repair pathway. These specialized bifunctional DNA glycosylases remove the 5-methylcytosine base and then clea...

Opposing roles for DNA structure-specific proteins Rad1, Msh2, Msh3, and Sgs1 in yeast gene targeting.

Targeted gene replacement (TGR) in yeast and mammalian cells is initiated by the two free ends of the linear targeting molecule, which invade their respective homologous sequences in the chromosome, leading to replacement of the targeted locus with a selectable gene from the targeting DNA. To study the postinvasion steps in recombination, we examined the effects of DNA structure-specific protei...
